Not sure what the "nature of the interfaces" means, but there are some tangible differences - lower latency (although the 2nd gen drivers work to an extent on the 1st gen units as well), more even gain structure (the 1st gen units' gain range tended to be bundled towards the loud end), higher maximum sample rate, surge protection, new instrument input with more headroom.
are you sure, you have current 2nd. generation of 6i6. Original Scarlett 6i6 has different drivers and software called Mix Control. Visually there is slightly different front panel and volume control knob is more "shiny" with black notch at 2nd. generation. Its driver is also available from the download section at Focusrite website.
